Packed Columns

CSPs designed for HPLC are widely used in SFC. This is because separations are performed at room temperature so that there is greater interaction energy between CSP-racemate (and therefore higher selectiv-ities) with fewer racemization problems. Packed-column SFC has the same advantages and this is why, since 1985, this technique has been successfully applied to chiral separations. However, the lack of commercial equipment for packed-column SFC has long been a major problem in the development of the technique; this handicap is now being overcome. The advantages of SFC over HPLC include: faster analysis, faster column equilibration, faster method development and also reduced generation of hazardous waste. It must also be underlined that SFC sometime exhibits thermodynamic advantages over HPLC by providing greater selectivity values (particularly with natural polymer CSPs).
