Offline, pre-column derivatizations have no ex-tracolumn loss of efficiency and no solvent or kinetic limitations. Derivatization can be conducted under flexible reaction conditions or with harsh reagents. Offline derivatization can be optimized for high reaction yields and minimum by-products. Derivatization solvents need to be miscible with the chromato-graphic mobile phase. Otherwise, the derivatization solvents have to be evaporated and the residue of derivatives redissolved in a solvent compatible with the mobile phase. Offline derivatization does not need to give 100% theoretical yields, as long as there is good sample-to-sample reproducibility. However, nonautomated offline pre-column derivatizations require operator attendance and manual manipulations.
