Ivermectin

Ivermectin in tissue is analysed with an ODS column using a mobile phase of acetonitrile-water (9:1) at 65°C. Ivermectin is detected fluorometrically at 272 nm excitation, 465 nm emission, with a detection limit of 0.25 ngg-1. Tissue sample is loaded onto a C8 solid-phase extraction column, eluted with acetonitrile, and the eluate dried under a stream of N2. The dried residue is dissolved with ethy1 acetate-hexane (2 : 3), loaded on a silica column, and eluted with methanol-ethyl acetate (1 : 1). The eluate is dried under a stream of N2 and treated with trifluoroacetic anhydride and methyl-imidazole. The analyte thus obtained is injected into an HPLC.
