Since its introduction by Porath in 1975, immobilized metal ion affinity chromatography (IMAC) has developed into a robust and versatile tool. The number of uses is large and includes the isolation of metal-binding compounds from sea water, separation of enantiomeric forms of amino acids, tetracycline removal from animal products and protein purification. This article will focus on its application to protein purification, where it relies on the ability of certain amino acid side chains to form coordinative interactions with immobilized metal ion chelate complexes. As a chromatographic method it falls somewhere between biospecific affinity chromato-graphy and ion exchange chromatography. The evolution of the technique, current tools and some specific technical details are discussed.
