Gentamycin

Gentamycin in plasma is extracted with a cation exchange solid-phase extraction column (carboxy-

propyl-bonded silica column). Gentamycin is eluted with a 1 : 1 mixture of acetonitrile-0.2 mol L_1 borate buffer (pH 10.5) and is derivatized with 9-fluorenylmethyl chloroformate. Derivatized gen-tamycin is analysed on an ODS column with a mobile phase consisting of acetonitrile-water (9 : 1). The derivatives are detected fluorometrically at 260 nm excitation, 315 nm emission, with a detection limit of less than 50ngmL~\ Gentamycins C1; C1a and C2 are resolved (Figure 1, structures 2, 3 and 4, respectively).

o-Phthaldialdehyde, dansyl chloride, fluorescamine, 1-fluoro-2,4-dinitrobenzene or 2,4,6-trinitroben-zenesulfonic acid may also be used as derivatizing reagents.
