Ferritin From Iron Storage to Nanoparticle Synthesis

Mineral deposition in the iron storage protein (ferritin) may be regarded as an archetypal biological model for the formation of a nanocrystalline mineral phase within a confined space. The structure and function of ferritin have been reviewed in great detail. Ferritin consists of an oligomeric protein shell (apoferritin) and a core of poorly crystalline Fe(III) oxyhydroxide (presumably ferrihydrite, 5 Fe2O3 • 9 H2O). Iron is temporarily stored within and released from the central cavity of the encapsulating protein shell. The availability of several high resolution three-dimensional structures of apoferritins originating from different organisms provides a reliable basis to discuss possible pathways of iron biomineral-ization. Current biomimetic strategies to achieve similar properties include mineralization in oil-water microemulsions, block copolymer micelles, or biotechnologically produced capsule-forming proteins.
